Coil Spilt hum cancel with single coil

I have a strat HSS with Alnico II Pro singles (RWRP in middle) and a 59 in the bridge. If I split the bridge humbucker, should I use the North (slug) or South (adj) coil with the RWRP single?

Depends on the polarity of the RWRP.

Ya. Standard Duncan singles are souf, rwrps are north.

My suggestion: swap the s/c pickup positions so the RW/RP is in the neck position, and the standard wound version in the middle.

Then use the standard Duncan split coil wiring, that splits to the Slug coil of the '59 humbucker.
