You might think about some Cool Rails or Hot Stacks for neck and mid depending on what kind of tone your going for.

In answer to the First question, Yes they do work together.. I have a L'il demon in the bridge and Duckbucker in Mid and Neck... I wouldn't recommend getting a Duckbucker for the Middle because i Think i'm going to go for a cool rail in the Mid because the Ducks cannot handle High gain, They're a low output pickup but they sound excellent clean(like an acoustic guitar)
I'm also thinking of changing my Demon to Hot Rails because IMO the l'il demon is too bright in strat types (light woods) so unless you have a heavy wooded strat e.g mahogany.. i'd suggest something else bar the l'il Demon....
That said though i still love the tone of the demon so it's personal taste...

The lilSD and the lil59 are highly complimentary.
Mid pickups aren't nessessary but they sure do make the middle 3 positions of a Strat possible. My Strat currently has a JB/Hot Stack/lil59 in it. The Hot Stack sounds very nice both alone and combined with the lil59. The JB is a little overpowering for a traditional Strat tone form that combo but I'm quite sure you'd get great results with the lilSD combined with the Hot Stack.
If you want to run the lilSD and lil59 at the same time, there are ways to wire that.
