If I were to install a hum-cancelling Hot rails pickup in the middle Strat position, a=along with a normal wound non hum cancelling neck and middel pickup, would the 2-3 and 4-5 positions be hum -cancelling?

To get hum cancelling, both pickups would have to be either double or single coil, not one of each. And if you are using 2 singles, one would have to be reverse wound/reverse polarity. But a single Hot Rails in a Strat can really best be useful in the bridge position.

You can use a super switch and split the rails in positions 2/4 to get hum cancelling. You'd have to work out magnetic and electrical polarity, but it should be doable.
