Is it possible? Can I use a multimeter to determine pot values in wired tone circuit in the guitar without unwiring it?

Yes, you can measure pots that are in a guitar.

Actually, in a tone circuit, it will give the exact value. The cap blocks the DC of the meter from going beyond the pot. It's one of the very few situations, in all of electronics, where you can get an accurate reading in-circuit.
