I don't see it interfering with your playing or improvising in the slightest. E flat tuning never bothered me, nor did dropped D.
Microtuning is another matter though. I tuned my bands to A-432 for years and I do believe it made a subtle difference. But it also screwed with my sense of vocal pitch a bit. I became so immersed in it that what I naturally heard in my head as an E didn't match what most music calls an E. When you have good pitch it's inconvenient being slightly out of tune with the rest of the musical world.
That never affected my guitar playing at all though.
Even so, I went back to 440 after a couple of years.
