The 5U4 will draw 3 Amps of filament current, the 5Y3 draws only 2 Amps. So you need to know the rating of the 5 Volt winding on your power transformer, it may well be only rated for 2 Amps, in which case it is not a good idea. There's also the DC Voltage drop to consider.

If it's an original Fender, I wouldn't even consider it unless I was going to install a new power transformer with the right spec's.

try a 5v4 if you want something a little hotter than a 5y3. same current draw and more power/less sag
