It definitely does.

A microphone won't hear the bass boost you get from coupling, so keep that in mind when dialing in the amp.

Sure makes a difference when moving an Ampeg 8x10.

My amp is so light (less than 25 pounds) it doesn't need casters.

I have the Ernie Ball pop-out casters on everything.

It's best to have the option to easily go on or off. Depends on the room and especially the floor construction. If the floor or room does not have too much rumble then I would almost always take the wheels off.

Its all carpet

Carpet will dampen highs but won't really have much affect on the lows. The floor material itself and the space under the floor will have a big affect on the rumble.
