For one, it's light. Most important, IMO, it helps vintage wind single coils ring more true. I use Fralin Vintage Hots in my strats and together with a light swamp ash body and a thin nitro finish you get the best strat tone (again, IMO). Think "Lenny" and "Riviera Paradise" and you get what I believe is the ultimate strat tone.
I have had heavier strats and there is no p/u that can help them. Slap that polyurythane crap all over it and you can forget about vintage tone. Now, if you are putting hot rails or something in the neck and middle, it doesn't matter. If you want a big fat sound from a strat, it comes from a good Fender-type amp, not the guitar. Then again, if you want a big fat humbucker tone, that's another story--get a LP.

Get ash or korina. Alder will give you a normal strat sound, but I prefer ash to alder... it's less chewy, more ballanced. I'd get mahogany before basswood, since basswood just gives a heavier sound than alder and mahogany does it better, but if you want that you might as well get korina. Korina and is similiar to mahogany but more resonant and less thumpy. I don't have experience with koa, but I've heard that it's like korina.

Go for an original strat wood config and select a good warm pup for the bridge and you should be doing just fine. Go with all Alnico 2 pickups (such as the custom bridge and APII in the neck and mid). Maple neck and Ash body aren't a bad way to go with the A2 pups either so they would deliver a nice snap but still have a balanced warmth.
