Does Strat Boat Routing Affect Tone?

Would boat routing (one large block carved connected to control cavity carved out for pickups as opposed to sss routing) it have a tonal effect of something like chambering perhaps?

If you're changing pickups in the guitar after rerouting it, you won't be able to notice the differences. I didn't when I had it done on my Stratocaster.

Minimal, IMO. Just don't go for a 1 ply guard if you route.

IMO, hell no. I have said this before, and here it is again: Three of the best sounding Strat-type guitars I've ever played have had swimming pool routs. Those are my two G&L Legacies and my Godin G-4000. That said, I also don't think there is any tonal benefit to a swimming pool rout. Don't bother permanently modifying the guitar if you don't have to in order to fit your particular pickups, in other words.

Some mass is removed so I think it would have a *tiny* affect on tone.

Completely unnoticeable though.

Negligible effect, IMO.

That being said, I prefer a HSH route because I hate the way a thinner or poor quality pickguard will sag into a swimming-pool route under the weight of two high-output humbuckers.

IMO, yes, on the basis that you can tell a difference between the two unplugged (particularly with a cheap pickguard, as mentioned).

Is it a huge difference? No. Would you notice if you were putting the same pickups back in? Very probably.

Is that difference one you would like? There's only one way to tell, and only one opinion that matters.

I'm in the boat (hehe) that it matters a lot.

Where "a lot" means noticeable without other changes. I did the test recording a Tele with original routes and enlarged to humbuckers, even that was showing up.

If I jab a Preparation H suppository up the jack hole of my Strat, will it tighten up my bass?

It's already boat routed, but I have an identical G&L which is routed HSH and only slightly heavier on the scale. To me, the boat route has a little more clarity, slightly more balanced mids, and a little louder acoustically when not plugged in. The wood grain looks very similar and so is the weight. They both have the same pickup (for now). It's hardly a difference, but I started noticing it over time.

Well I wouldn't know, but your sister tells me that your little pecker will fit in an 1/4" jack. I wouldn't suggest trying that either.

Identical. like made with EXACTLY the same wood....as in you pulled it out of a parallel universe.

Not even guitars made with lumber from the same tree will have identical properties.

You have different guitars - yes they will sound different.

Also the strength of the guitar decreases IMHO. If ever it takes a header, i think the non-pool routed one will keep its shape easier than the pool routed. BTW, one of Strats' advantage is their durability in case of mal-usage, accidents, etc... (in comparison to angled headstock, set-in neck designs)

^^ I doubt that. After all there's that cavity for the wiring running from neck PU cavity to bridge pu cavity. If there's a difference in strength, its slight.
