ANYthing in or on a guitar makes string vibration a variable quality . EVERYTHING effects tone. Rubber sounds diff from springs - Bakelite teles do sound unique inmho.

Do you mean 'does wood affect tone'

or

Do pickups mounted on a pickguard suffer a tone loss (whatever that is) based on what wood the pickguard is screwed to
