I buy pretty much all of my stuff at the same local shop because they beat web deals for friends and have great service so I can generally try the guitar through the same model as the amp I have. First I just look it over, eye the fretwork and binding and the nut. Then I feel the neck joint and play a song I know really well unplugged to look for fret buzz. If there is any of if the action is a bit off then I try to visualy figure out if I could easily correct it. Then I play it through a few different amps in many settings for quite a while. Then I play every other promising guitar in the store through the same amps to get an idea of the tone. If I like it I buy it. Don't go to a store to buy unless you have many hours to just play everything in the store. Make sure to compare it to a cheap guitar and the most expensive one in the shop to get an idea where it is on the food chain so to speak.
