I have guitars with pickups that complement each other well and I know how to work the volume and tone controls on my guitar.

i set the amp so that it brings out the qualities of the pickups i use .... i go for a lot of versatility, so the amp has to be able to bring about all of the tones in a balanced way ... so i tweak alot (a little on the neck - a little on the bridge - a little with the combinations - etc) to get it set so that the pickup selector ends up producing the range i am going for ... my biggest issue has been volume balance between full on and split tones ... the 'solo' boost on the mesa helps with this alot ... i set the amount of boost so that the single and split tones are as loud as the full humbucker tones

I EQ for the bridge pup too. I mostly play using the bridge pup. I tend to use the guitars vol and tone knobs to dial in the right neck tone, while leaving the bridge pup fully open.

I eq for the bridge, too, too, and the neck pup has tone either on 10 or 5-7 for soloing/cleans with vol. rolled off to ~1,3. (LP w/ Custom/PG)

Bridge for all distorted tones.

If it sounds great on the bridge pickup but not on the neck pickup... time for a new neck pickup.

That is my exact approach also, I match the pups for what I want. Though I do use one channel on the Road King specifically for strat neck sounds.

My neck pickup sounds like ass no matter how I EQ the amp, so I just worry about the sound of the JB. :D

I use my bridge pickup about 95% of the time, so I definitely start there. I tend to choose guitars and neck pickups that perform satisfactorily using that approach.

A combination... Generally when my bridge sounds good, so does my middle and neck. :)

On my G&L Legacys, I set the amp up for the bridge clean tone. I want the maximum amount of treble that I need, without being harsh; for a spanky, country twangy guitar tone. The G&Ls have global passive Treble and Bass controls, so I can get just about anything I want from here.

With a Les Paul, I try to go for a good, broad acoustic guitar-like rhythm tone from the middle switch position (both pickups on) with all controls full up, then I adjust the controls as needed.

I use Mesa Boogie amps, so I always have excellent control over my LEAD Channel EQ.
