Does a 4x12 have more clean headroom than a 2x12? And if so, is it significantly more?

Well, much depends on the speakers used, but:
let's say you have a 2x12 and a 4x12 loaded with identical speakers, running the same wattage.
when you couple two speakers together in a cabinet, there is a coupling effect that is the result of those two speakers being mounted on the same baffle that raises the overall output by a few decibels when compared to a single speaker. Add two more and the effect increases.
So, yeah, a quad will put out a louder clean than a duo bin with identical speakers. And it will be louder overall given the same amp input.
