Does this guitar have a Floyd Rose or similar bridge? Sounds like the bridge is being pulled down, you may have to do a trem spring adjustment to balance the tension that is lacking because of the de-tune.........Joe

how high is the string height at the 12th fret when all strings are open? I generally stick a 2mm pick between the fret wire and the string
it should either stick lightly or pass slightly. if that happens your bridge is high enough and you have other problems. but you might also want to look at making sure your trem is "floating" the plate all the saddles sit on should be level and parallel with the body surface. adjust that by adjusting the springs in the trem cavity. you may need to play with that for a long while before the strings are tuned properly and the trem is level. Floyd's are a pain to setup but theyre worth the time and effort. once you have the bridge height adjusted and the strings are 2mm off the fret wire at the 12th fret when all strings are open you can then start to fine tune your intonation and truss rod. ultimately you want the strings to be about level across the fretboard. of course they wont be totally level but it should be close. you dont want a huge rise in the strings as it goes down the neck.

I am using whatever came on the guitar. I would say 10-52 maybe?

I ultimately want to go up to 11-70 zakk wyldes for detuning on this one

If you hold down the 14th fret, the bridge height has nothing to do with it any more. Think about it...

It looks to me like the nut is turning by itself and the truss rod isn't actually moving. Not sure what to do about it, but there's no need to panic.

try those 11-70 ZWs and see what happens.
