Is that 'high gain amp noise' a whistle or feedback of some sort? That combined with the 'dusty' sound you describe could indicate a microphonic preamp tube. Take the back panel off of the amp and tap on your preamp tubes with a pencil or similar. You may hear a faint click if the tubes are working properly, whereas a microphonic tube will produce much louder noise.

Do I do this just when my guitar is plugged in? In On mode?

You don't need to have your guitar plugged in, but the amp needs to be operating. You may want to let it warm up for several minutes to be sure the tubes are sufficiently warm before taking it off standby.

In the end...tapping on tubes (which is not good for them anyway) does not tell you much. It might identify a tube that is Very Microphonic.
But do not let tapping be your acid test.
The only way to know if a tube is bad in an amp you are playing, is to pull said tube, and install a replacement.
You can even switch a tube that is "bad" in V1, and have success with it in another slot.
I always thought that was inviting future problems.....but whatever.

So that being said, it could be more of an amp problem than tube problem ya?
