Thanks for the responses everyone... with so much talk about wood and glue and minor changes in tone it was something that was on my mind when thinking about strats that have large pickup routes that sit empty.

Any filling would do nothing.

Having wood unrouted and/or no pickup would potentially make a difference. Construction certainly changes nuances in tone. Those who have esquire teles and single p90 juniors feel those guitars are raunchier. Maybe there's something in having more unadulterated wood near the neck join.
